Assessing the effects of urbanization on streams in Tierra del Fuego Ecología austral
Zagarola,Jean-Paul A; Martínez Pasteur,Guillermo; Lopez,María Eugenia; Anderson,Christopher B.
We set out to understand how urbanization affects streams in Ushuaia, Tierra del Fuego. Paired t-tests and linear regressions were used to compare physico-chemical stream habitat variables (i.e., temperature, turbidity, conductivity, dissolved oxygen and pH) and benthic macroinvertebrate community structure (i.e., density, taxonomic richness, Shannon-Weiner diversity) and function (functional feeding groups [FFG]) in four watersheds with urban and reference sites. We then calculated indices of biotic integrity for habitat (rapid visual assessment protocol [RVAP]) and benthos (Ephemeroptera, Plecoptera, Trichoptera richness [EPT], family biotic index [FBI], rapid bioassessment protocol [RBP] and biotic monitoring Patagonian streams [BMPS]). Results...

Genotoxicity evaluation of domestic sewage in a municipal wastewater treatment plant Genet. Mol. Biol.
Grisolia,Cesar Koppe; Oliveira,Adriano Barros Barata de; Bonfim,Helder; Klautau-Guimarães,Maria de Nazaré.
An evaluation of the genotoxicity of domestic sewage was carried out in a municipal wastewater treatment plant. The study was carried out using Allium cepa root tip cells through cytological parameters such as aberrant cells in anaphase-telophase and the mitotic index, and the fish species Oreochromis niloticus and Tilapia rendalli, through the index of micronuclei in peripheral erythrocytes. In the Allium test, each of the four stages of the wasterwater treatment routine was analyzed, i.e., crude sewage, primary effluent, secondary effluent, and tertiary effluent. O. niloticus and T. rendalli specimens were maintained in an aquarium continuously receiving the final effluent, in an open system, before being discharged into Lake Paranoá. For micronuclei...

Comet assay comparison of different Corbicula fluminea (Mollusca) tissues for the detection of genotoxicity Genet. Mol. Biol.
Rigonato,Janaina; Mantovani,Mário Sérgio; Jordão,Berenice Quinzani.
The comet assay was used to study the sensitivity of the widely distributed freshwater bivalve mollusk Corbicula fluminea to the DNA-damaging alkylating-agent methylmethane sulfonate (MMS). This study was undertaken to ascertain if C. fluminea is a good bioindicator of pollutants in aquatic environments and identify which C. fluminea tissue is most effective and practical for genotoxicity studies. The mollusks were exposed to 0.6, 1.2 or 2.4 X 10-4 M MMS for 40 min and their hemolymph, gill tissue and digestive gland tissue assessed for the level of DNA damage and the time needed for the tissues to recovery. Regression analysis showed a direct linear dose-response relationship between MMS concentration and the number of damaged cells for hemolymph and...

Profile of micronucleus frequencies and nuclear abnormalities in different species of electric fishes (Gymnotiformes) from the Eastern Amazon Genet. Mol. Biol.
Melo,Karina Motta; Alves,Ingrid Reale; Pieczarka,Julio Cesar; David,José Augusto de Oliveira; Nagamachi,Cleusa Yoshiko; Grisolia,Cesar Koppe.
The frequency of spontaneous micronucleus (MN) formation in fish species needs to be determined to evaluate their usefulness for genotoxic biomonitoring. The definition of a good bioindicator takes into account the current knowledge of its metabolic traits as well as other factors including its feeding behavior and relationship to the environment. In this study, we compared the basal frequencies of micronucleated erythrocytes and nuclear abnormalities (NA) among different species of the fish Order Gymnotiformes (Rhamphichthys marmoratus, Steatogenys elegans, Sternopygus macrurus, Parapteronotus hasemani, Gymnotus mamiraua, Gymnotus arapaima, Brachyhypopomus beebei, Brachyhypopomus n. sp. First genotoxicity study of Paraná river water from Argentina using cells from the clam Corbicula fluminea (Veneroida Corbiculidae) and Chinese hamster (Cricetulus griseus Rodentia, Cricetidae) K1 cells in the comet assay Genet. Mol. Biol.
Caffetti,Jacqueline D.; Mantovani,Mário S.; Pastori,María C.; Fenocchio,Alberto S..
High concentrations of xenobiotics from urban and industrial wastes have contributed to the contamination of many aquatic environments. We used the comet assay to evaluate the genotoxic potential of water collected from the River Paraná, which receives a great deal of waste, at three points (Puerto Piray, Eldorado and Montecarlo) in the Misiones Province of Argentina. The in vivo comet assay used 40 freshwater clams (Corbicula fluminea) while the in vitro comet assay used Chinese hamster (Cricetulus griseus) K1 cell (CHO-K1) cultures with the mutagen ethyl methanesulfonate (EMS) as the positive control and phosphate buffered saline (PBS) as the negative control. Both assays showed statistically significant differences between the three sampling sites in...

Etude des variations saisonnières de différents biomarqueurs, chez la moule Mytilus galloprovincialis. Relation avec certains paramètres physiques, chimiques et physiologiques ArchiMer
Bodin, Nathalie.
The aim of this study was to evaluate the seasonal variations of six biomarkers on Mytilus galloprovincialis, using comparison with physiological index and chemical contamination levels on two sites of  Mediterranean sea. The seasonal variations of acetylcholinesterase (AChE), DNA adducts, benzo(a)pyrene hydroxylase (BPH), heat-shock proteins (HSP), metallothioneins (MT) and multixeniobiotic proteins (MXR) were estimated over a two-year period, on mussels which were sampled in Carteau (original site) and La Fourcade (transplanted organisms). Carteau, an industrial site, presents higher contamination levels (HAP, PCB, heavy metals) than La Fourcade, an urban area. The PAH profiles underline a strong spatio-temporal variability of levels and sources of...

Sampling design optimization for EROD measurements in fish ArchiMer
Beliaeff, Benoit; Burgeot, Thierry.
Activity of 7-ethoxyresorufin-O-deethylase (EROD) in fish is certainly the best-studied biomarker of exposure applied in the field to evaluate biological effects of contamination in the marine environment. Since 1991, a feasibility study for a monitoring network using this biomarker of exposure has been conducted along French coasts. Using data obtained during several cruises, this study aims to determine the number of fish required to detect a given difference between 2 mean EROD activities, i.e. to achieve an a priori fixed statistical power (l-beta) given significance level (alpha), variance estimations and projected ratio of unequal sample sizes (k). Mean EROD activity and standard error were estimated at each of 82 sampling stations. The...

Our planet is changing, and one of the most pressing challenges facing the scientific community revolves around understanding how ecological communities respond to global changes. From coastal to deep-sea ecosystems, ecologists are exploring new areas of research to find model organisms that help predict the future of life on our planet. Among the different categories of organisms, meiofauna offer several advantages for the study of marine benthic ecosystems. This paper reviews the advances in the study of meiofauna with regard to climate change and anthropogenic impacts. Four taxonomic groups are valuable for predicting global changes: foraminifers (especially calcareous forms), nematodes, copepods and ostracods. Environmental variables are fundamental in...

Modélisation de la bioaccumulation de métaux traces (Hg, Cd, Pb, Cu et Zn) chez la moule, Mytilus galloprovincialis, en milieu méditerranéen ArchiMer
Casas, Stellio.
Monitoring coastal contamination of trace metals using bivalve molluscs as quantitative bioindicators is widely used in many international monitoring programs. The bioaccumulation of metal within an organism results from the interaction between physiological (growth, loss of weight, absorption, accumulation), chemical (metal concentration, speciation, bioavailability) and environmental factors (temperature, SPM, chlorophyll). By utilizing the parameters above in this study, it appears that modeling metal dynamics in marine mussels is a reliable tool. The model allows understanding the bioaccumulation process which results from the interactions between biological, chemical and environmental factors. One of the objective of this study was to couple a...

Environmental DNA (eDNA) analysis is a revolutionary method to monitor marine biodiversity from animal DNA traces. Examining the capacity of eDNA to provide accurate biodiversity measures in species‐rich ecosystems such as coral reefs is a prerequisite for their application in long‐term monitoring. Here, we surveyed two Colombian tropical marine reefs, the island of Providencia and Gayraca Bay near Santa Marta, using eDNA and underwater visual census (UVC) methods. We collected a large quantity of surface water (30 L per filter) above the reefs and applied a metabarcoding protocol using three different primer sets targeting the 12S mitochondrial DNA, which are specific to the vertebrates Actinopterygii and Elasmobranchii. By assigning eDNA sequences to...

Towards an Autonomous Pelagic Observatory: Experiences from Monitoring Fish Communities around Drifting FADs ArchiMer
Brehmer, Patrice; Sancho, Gorka; Trygonis, Vasilis; Itano, David; Dalen, John; Fuchs, Ariel; Faraj, Abdelmalek; Taquet, Marc.
This work presents a methodological synthesis for the in situ monitoring of fish aggregating devices (FADs) using a combination of optical, echosounder and SCUBA observations conducted in the vicinity of drifting FADs. The acoustic methods allowed, according to the devices used, the description of the spatial organisation and dynamics of biotic scattering layers, individual fishes, schools, shoals and mammals, while visual, photographic and video observations permitted species identification within a range of 0 to ~ 25 m. Based on these results, we elaborate on the interest to combine acoustic and visual methods, and present an autonomous instrumented drifting buoy for remotely monitoring fish diversity and abundance in the pelagic ecosystems. The...

Utilisation des biomarqueurs pour la caractérisation de l’état écotoxicologique des masses d’eau ArchiMer
Sanchez, Wilfried; Porcher, J.-m..
In the past 25 years, numerous biomarkers have been developed with the objective to apply them to aquatic ecosystem biomonitoring. Recently, the Water Framework Directive (WFD) of the European union specified monitoring programs required to assess the achievement of good chemical and ecological status for all water bodies by 2015. This article reviews the potential of biomarkers for ecotoxicological status assessment in WFD monitoring programmes based on previous national and international experiments.

Long-term records of cadmium and silver contamination in sediments and oysters from the Gironde fluvial-estuarine continuum - Evidence of changing silver sources ArchiMer
Lanceleur, Laurent; Schaefer, Joerg; Chiffoleau, Jean-francois; Blanc, Gerard; Auger, Dominique; Renault, Sophie; Baudrimont, Magalie; Audry, Stephane.
The Gironde fluvial estuarine system is impacted by historic metal pollution (e.g. Cd, Zn, Hg) and oysters (Crassostrea gigas) from the estuary mouth have shown extremely high Cd concentrations for decades. Based on recent work (Chiffoleau et al., 2005) revealing anomalously high Ag concentrations (up to 65 mg kg(-1); dry weight) in Gironde oysters, we compared long-term (similar to 1955-2001) records of Ag and Cd concentrations in reservoir sediment with the respective concentrations in oysters collected between 1979 and 2010 to identify the origin and historical trend of the recently discovered Ag anomaly. Sediment cores from two reservoirs upstream and downstream from the main metal pollution source provided information on (i) geochemical background...

Temporal study of estrogenic responses of mussel (Mytilus galloprovinciallis) extracts applied to reporter cell lines ArchiMer
David, A; Dagnino, S; Pichot, Yves; Munaron, Dominique; Escande, A; Casellas, C; Fenet, H; Gomez, E.
In vitro bioassays would facilitate monitoring of estrogen-like compounds in mussels (Mytilus galloprovincialis) since they tend to accumulate lipophilic compounds in their fat and muscle tissue. However, estradiol (E2) steroid (already identified in mussels) could induce false positive responses. This study focuses on temporal variability in estrogenic responses of mussel extracts and the possible relation of this response with E2 levels. The reproductive cycle and growth were thus monitored for six months in sexed mussels. E2 levels were similar between genders and these levels varied with reproductive stages. human estrogen receptor (hER) activities were similar at all stages except in February. E2 present in male and female extracts accounted for part...

L'éthoxyrésorufine-o-déethylase, les adduits à l'ADN et les mocronuclei dans les organismes marins. Application à la surveillance des effets biologiques sur les côtes françaises ArchiMer
Burgeot, Thierry.
Three biomarkers [ethoxyresorufm-o-deethylase (EROD), DNA adducts and micronuclei (MN) were studied for biomonitoring applications along French coasts. EROD was developed at an experirnental stage in two pilot sites, Seine Bay and Fos sur Mer, using the dab (Limanda limanda), dragon et (Callionymus lyra), red mullet (Mullus barbatus) and comber (Serranus hepatus) as target species. Higbest EROD activity was observed in the subestuarial part of Seine Bay and at the stations closest to sources of pollution from Cortiou and Fos sur Mer. The suitable number of fish for detection of a significant difference between two EROD activities was deterrnined as a function of sampling variance and a preselected statistical power. Lead contamination of small cetaceans in European waters - The use of stable isotopes for identifying the sources of lead exposure ArchiMer
Caurant, Florence; Aubail, A; Lahaye, V; Van Canneyt, O; Rogan, E; Lopez, A; Addink, M; Churlaud, C; Robert, M; Bustamante, Paco.
Lead concentrations and isotopic composition have been measured in bone and teeth of small cetaceans belonging to three species (Delphinus delphis, Phocoena phocoena and Stenella coeruleoalba), to evaluate the toxicological risk and to determine sources of lead in the European waters. Lead concentrations, far lower than threshold value inducing toxic effects in human, were higher in teeth than in bones, but highly correlated between the two tissues (r = 0.92, p < 0.001). Large variations of Pb-206/Pb-207 values in bone tissue showed that cetaceans must be submitted to various atmospheric influences. No geographical differences appeared which is consistent with studies on their distribution indicating seasonal movements between Brittany waters and the...

To assess the effect of anthropogenic activities on ecosystems, it is of prime importance to develop new tools enabling a rapid characterization of ecological communities. Freshwater ecosystems are particularly impacted and threatened by human activities and need thorough attention to preserve their biodiversity and the ecological services they provide. Studying such ecosystems is generally difficult because the associated organisms are hard to sample and to monitor. We present a ready-to-use environmental metabarcoding protocol to characterize and monitor the freshwater gastropods communities from water samples. The efficiency of this new tool was compared to a classical malacological survey at 19 sampled sites from 10 distinct rivers distributed over...

Multixenobiotic resistance protein expression in Mytilus edulis, M-galloprovincialis and Crassostrea gigas from the French coasts ArchiMer
Minier, C; Moore, M; Galgani, Francois; Claisse, Didier.
Multixenobiotic resistance (MXR) is a membrane-transport mechanism that allows organisms to exclude many compounds from their cells and tissues. It is thus a first line of defence against a variety of toxic compounds. Since mussels and oysters possess MXR proteins, an analysis of the expression level of these membrane-transporters has been conducted in relation to their body burden of some major environmental contaminants. Mussels Mytilus edulis and M. galloprovincialis and the oyster Crassostrea gigas were sampled from a total of 43 sites along the French coasts. High expression levels were found in animals from the major French estuaries (Seine, Loire and Gironde), at a few sites in Brittany and in nearly all sites from the Mediterranean mainland coasts....
